Just to keep it simple:
DOS palette has a few extra colors, so artists should prefer that. OTTD prefers DOS palette for the same reason.
And don't forget: In 32bpp you do not need any palette unless you will use color cycling and company colors.

could i draw a sprite in RPB not using the palette when apply the Dos palette which converts the colours to match?

would that work?

Would work, but the colors might look horrible. I have done it and replaced those that didn't look OK-but my primary intention was to draw 32bpp and then provide the 8bpp too. ;-)
